小结
文章中有多个补gap软件,如TGS-gapcloser + LR_Gapcloser;还有利用minimap2比对,winnowmap2比对,手动补gap的,还有对特定区域PCR补gap,还有利用其他组装软件组装的基因组补洞,比如nextdenovo,verkko等等。TGS-gapcloser 不同ont pb参数,minimap2比对参数,多尝试效果会不同。
2.一般推荐使用TGS-GapCloser+ quarTeT进行补洞,分别使用三代数据和其他组装contig序列(nextdenovo,verkko等)进行补洞,,得到最终的T2T基因组。
3.总之不同软件效果不太一样,比如序列,长度都不太一样,选择能合理解释的结果即可,也可以结合多个软件补完的结果。
4.补完之后最好能加个验证。
